چکیده

The tumor promoter 12-O-tetradecanoylphorbol-13-acetate (TPA) rapidly blocks the terminal differentiation of normal cul tured Chondroblasts. In TPA, the chondroblasts lose their char acteristic polygonal morphology and initiate the synthesis of an atypical type IV sulfated proteoglycan. This atypical proteoglycan has lower molecular size, shorter polysaccharide chains, and different 4S-disaccharide:6S-disaccharide ratio in com parison with type IV sulfated proteoglycan from untreated chon droblasts. When removed from TPA, the cells reacquire their characteristic polygonal morphology and reinitiate the synthe sis of their typical cartilage-characteristic type IV sulfated pro teoglycan. TPA has no readily detectable effect on the type III sulfated proteoglycan synthesized by fibroblasts.

Transforming Growth Factor-β1 Preserves Bovine Nasal Cartilage against Degradation Induced by Interleukin-1α in Explant Culture

Background and Aims: Chondrocytes and their differentiation play a central role in joint diseases. Effect of the transforming growth factor (TGF)-β1 on chondrocyte characteristics and differentiation is not clearly understood. This study was undertaken to investigate the effects of TGF-β1 on tissue characteristics and morphology of chondrocytes against degradation induced by interleuk...

Effects of phorbol-12-myristate-13-acetate on the phenotypic program of cultured chondroblasts and fibroblasts.

Phorbol-12-myristate-13-acetate (PMA) has a prompt, differential, and partially reversible effect on cultured chick chondroblasts. Within 36 hr PMA transforms sessile, polygonal, epithelioid chondroblasts into motile, multilayered, fibroblastic cells. In PMA chick chondroblasts rapidly cease to synthesize two of their terminal luxury molecules, the type IV sulfated proteoglycan that characteriz...

Enhanced synthesis of a specific protein by 12-O-tetradecanoylphorbol-13-acetate in cultured chick embryo muscle cells.

The tumor promoter 12-O-tetradecanoylphorbol-13-acetate (TPA) induces in cultured postmitotic myotubes specific alterations of synthesized proteins as revealed by one-dimensional sodium dodecyl sulfate-polyacrylamide gel electrophoresis after pulse labeling with [35S]methionine. Synthesis of myosin heavy chain is remarkably inhibited after exposure to 1.6 X 10(-7) M TPA for periods of 9 hr or l...

Cellular transformation and differentiation. Effect of Rous sarcoma virus transformation on sulfated proteoglycan synthesis by chicken chondrocytes.

Incorporation of sulfate into sulfated proteoglycans by isolated chicken chondrocytes was inhibited up to 74% by transformation with the Rous sarcoma virus, and a similar inhibitory effect was observed on acetate incorporation into chondroitin sulfate. Slower sedimenting sulfated proteoglycans appear after the viral transformation. The ratio of chondroitin 4-sulfate to chondroitin 6-sulfate in ...
